I sometimes watch movies and shows with my brother using module-combine-sink so that we can listen to the audio on multiple headphones. However, with JamesDSP running, I get three sinks (one additional created by JamesDSP). How do I configure it with module-combine-sink so that I can get the output on both the headphones?

asuming you have pipewire
you use Q-Jacktctl and connect both outputs to jamesdsp_filter (I believe its called like that).
IMPORTANT: You have to playback sth so that jdsp_filter is shown.

you do it with qjack and not with module-combine-sink
(other tooling)
